Chapter 13, Problem 15P

Two yeast strains were mated and sporulated (allowed to carry out meiosis). One strain was a haploid with normal chromosomes and the linked genetic markers ura3 (requires uracil for growth) and arg9 (requires arginine for growth) surrounding their centromeres. The other strain was wild type for the two markers (URA3 and ARG9) but had an inversion in this region of the chromosome as shown here in pink:

Chapter 13, Problem 15P, Two yeast strains were mated and sporulated allowed to carry out meiosis. One strain was a haploid

During meiosis, several different kinds of crossover events could occur. For each of the following events, give the genotype and phenotype of the resulting four haploid spores. Assume that any chromosomal deficiencies are lethal in haploid yeast. Do not consider crossovers between sister chromatids.

a. A single crossover outside the inverted region
b. A single crossover between URA3 and the centromere
c. A double crossover involving the same two chromatids each time, where one crossover occurs between URA3 and the centromere and the other occurs between ARG9 and the centromere

Here is a depiction of the position of several genes on 2 chromosomes, with a dash "-" depicting the position of the centromere. Chromosome A has genes ABCD-EFGHIJ Chromosome B has genes 1234-56789. Please name the kind of rearrangement that would result in the following derivative chromosomes: 12D-EFGHIJ AB4-56789 O Unbalanced reciprocal translocation Unbalanced nonreciprocal translocation Balanced reciprocal translocation Balanced nonreciprocal translocation
The STR DNA marker DS11 is located on the p arm of the chromosome #8 in humans. Molecular analysis has shown that Charlie Chaplin is a heterozygote for the STR marker (genotype DS11A and DS11B) Using this knowledge, provide a diagram of meiosis showing only the movements of chromosome 8 (assume no nondisjunction). Throughout, indicate where the marker may be found (in other words, label the chromosome to show where the STR markers can be found. (Show ONLY the chromosomes at interphase, metaphase 1, end of meiosis 1, metaphase 2, and end of meiosis 2). Suppose Charlie conceived a son with a chromosome 8 trisomy. Molecular analysis of the STR DS11 marker revealed that his son carried two copies of the DS11A marker and a DS11C marker. Based on the data, determine if the nondisjunction occurred in Charlie or in the mother?  Provide your reasoning.
Colchicine is a chemical mutagen that inhibits the spindle formation and prevents anaphase, which retains the cell’s single restitution nucleus (doubled chromosome number). Suppose that an onion (2n=16) is subjected to three consecutive rounds of colchicine treatment, what will be the resulting chromosome number of the treated onion?
