Concept explainers
To review:
The expected result of the blot (a, b, or c) based upon the information about ferritin mRNA (messenger ribonucleic acid) regulation and the expected result if, the gel is probed with complementary DNA (deoxyribonucleic acid) of transferrin receptor mRNA.
Introduction:
The Northern blotting technique is used to identify the type and amount of RNA that isexpressed in a cell. The RNAs that are isolated from the cells are separated by gel electrophoresis which, is then blotted to a nylon membrane. The membrane is placed in a solution of a labeled DNA probe which, is complementary to a particular RNA. The bands thus formed, are analyzed to determine the type of RNA complementary to probed DNA and the position or thickness of the bands reveal the amount of RNA that is expressed in a particular cell.

Genetics: Analysis and Principles
- Some S. aureus strains can produce an enzyme called beta-lactamase, which breaks down beta-lactam drugs like methicillin. These strains that make beta-lactamase have a gene called mecA. Draw a diagram of the bacterial DNA encoding this gene, including the following terms: promoter, terminator, coding region.arrow_forwardDuring experimental RNAi, how does the researcher affect expression of a target gene? Group of answer choices Inject custom, lab-made double-stranded RNA into the specimen that will be targeted by the cell's existing Dicer to create siRNA complementary to the target gene's transcript. Inject lab-made Dicer proteins into the specimen which will cut up the target gene's transcript. Inject custom, lab-made siRNAs complementary to the target gene's transcript.arrow_forwardWhat is homologous recombination and where/when does it typically take place in an organism. Homologous recombination is also used experimentally during gene targeting. If you wanted to remove the function of the mouse gene fibroblast growth factor 4 (FGF4) using gene targeting by homologous recombination, what are the different aspects of a targeting vector you would need to make?arrow_forward
- Mouse genomic DNA is treated with a restriction endonuclease and electrophoresed in an agarose gel. A radioactive probe made from the human gene rxr-1 is used to perform a Southern blot. The experiment was repeated three times. Explain the results of these repeated experiments:arrow_forwardBacteria exposed to viruses incorporate sections of the virus’s DNA into the CRISPR array sequences in their genome. This mechanism allows bacteria to fight off the viruses, like an immune response: the information in CRISPR spacers served as “coordinates” for recognizing and cutting up invading DNA sequences. Describe what might happen under the conditions described after a bacteriophage infects a bacterial cell and releases its DNA into the bacterial cell. Explain why: The cas genes on the bacterial genome contains a frameshift deletion mutation that alters the function of the protein The bacteria will be unable to elicit an immune response and will succumb to the phase infectionarrow_forwardNorthern blots are valuable tools to analyze the mRNA level present in a sample. Describe an experiment that uses the Northern blot as a tool and how you would visualize the mRNA giving at least two examples of probe types.arrow_forward
- Bacteria can be used to produce human growth hormone (HGH - a peptide/protein) through genetic engineering. The human gene for HGH is inserted into a plasmid, which is then taken up by a bacterial cell, which divides and multiplies into a clone of cells, all of which contain the plasmid with the HGH gene. The bacteria express the HGH gene, producing HGH which can be harvested and used for treatment of humans. (See figure below) Which of the following statements is NOT true about this process? bacterium Vector, such as a DNA containing the gene of plasmid, isolated it from a different species is Gene encoding protein for pest resistance is inserted into plant cells ©2019 Pearson Education, Inc chromosome recombinant DNA (plasmid) transformed bacterium Create and harvest copies of a gene with either of two goals in mind. You can choose either a bacterial expression or yeast expression system. Which one would be a better option? Justify your choice.arrow_forwardResearchers have identified a gene (FR) responsible for watermelon resistance to infection by Dacus curcurbitae (a close relative of Drosophila melanogaster). They isolate RNA from resistant (FR+) and sensitive (fr-) watermelons and use a probe that will recognize both FR+ and fr- transcripts. They also isolate protein from resistant and sensitive watermelons and perform a Western blot using an antibody that can recognize the fr- and FR+ protein. Describe the results illustrated below and give a plausible molecular explanation for these observations.arrow_forward
