Chapter 29, Problem 11EQ
Summary Introduction
To review:
The criteria that could be used to establish the credibility of the dinosaur sequence extracted from the dinosaur bone.
Introduction:
Dinosaurs are an extinct group of reptiles. They first appeared in the Triassic period. At that time, they were dominant terrestrial vertebrates and ruled the land. Modern time birds and reptiles have been said to be derived from dinosaurs, with the course of evolution. The largest dinosaur of the 40 feet fossil was found to be that of Tyrannosaurus rex.

In 1979, bones found outside Ekaterinburg, Russia, were shown to be those of Tsar Nicholas and his family, who were executed in 1918 by a Bolshevik firing squad in the Russian Revolution . To prove that the skeletons were those of the royal family, mtDNA was extracted from the bone samples, amplified by PCR, and compared with mtDNA from living relatives of the tsar’s family.
Q. Mitochondrial DNA from which living relatives would provide useful information for verifying that the skeletons were those of the royal family?
How do you know if the halibut you purchased at the supermarket is really halibut? To identify the source of a biological sample, scientists PCR amplify and then sequence a region of DNA known to vary between species. For animals, this DNA region is a 648-base pair portion of the mitochondrial cytochrome oxidase I gene. The sequence of this mtDNA region acts as a so-called DNA barcode because a database exists that contains the sequences of this mtDNA region that are unique for hundreds of thousands of animal species.
A) Why do you think that a region of mitochondrial DNA is used for barcoding animals, as opposed to a region of nuclear genomic DNA?
A researcher used polymerase chain reaction (PCR) to test four individual nematodes
for the presence of a mutation in Gene Y.
When amplified, the fragments of DNA containing the mutated gene alleles were
250 base pairs long and the normal (wild type) alleles were 350 base pairs long.
To identify the genotypes of the individual nematodes, the researcher used gel
electrophoresis to visualize the PCR results.
1.000 bp
500 bp
300 bp
200 bp
100 bp
Remember that these nematodes are diploid organisms and as such, inherited two
copies of Gene Y.
Which of the following statements is a correct analysis of these results
a) Nematode As homozygous for this gene and has two copies of the mutated
allele.
O b) Nematode C is heterozygous for this gene and has one copy of the mutated
allele and one copy of the normal (wild type) allele.
O) Nematode D is heterozygous for this gene and has one copy of the mutated
allele and one copy of the normal (wild type) allele.
) d) Nematode B is homozygous for this…

- Amoebic keratitis is a rare eye infection associated with the use of soft contact lenses. The infection occurs when certain types of amoebas gain entry into the eye through the use of contaminated contact lenses. Medical researchers have used a restriction enzyme digest to characterize the type of amoeba found in patients with amoebic keratitis. These researchers extracted the mitochondrial DNA from infection-causing amoebas and digested, or cut, the DNA with a restriction enzyme. They found that, when visualized using DNA gel electrophoresis, the cut samples contained five bands of the following sizes: roughly 4,200 base pairs (bp), 5,500 bp, 6,500 bp, 9,300 bp, and 10,500 bp. To determine the prevalence of infection-causing amoebas in a certain region, researchers obtained samples from contact lens cases in a representative set of households. The researchers isolated the amoebas present in each sample, then used the protocol described above to view the DNA banding patterns of the…arrow_forwardGenetic analysis of Mycobacterium leprae, the bacterium that causes leprosy, reveals that its genome has undergone decay over time, losing DNA and acquiring mutations that make some of its genes nonfunctional.
