Chapter 6, Problem 6.8PS

QUANTITATIVE Turnover Number. Carbonic anhydrase catalyzes the reversible hydration of carbon dioxide to form bicarbonate ion:

CO 2 + H 2 O HCO 3 + H +     (6-19)

As we will discover in Chapter 8, this reaction is important in the transport of carbon dioxide from body tissues to the lungs by red blood cells. Carbonic anhydrase has a molecular weight of 30,000 and a turnover number (kcat value) of 1 × 106 sec−1. Assume that you are given 1 mL of a solution containing 2.0 μg of pure carbonic anhydrase.

  1. (a) At what rate (in millimoles of CO2 consumed per second) will you expect this reaction to proceed under optimal conditions?
  2. (b) Assuming standard temperature and pressure, how much CO2 is that in mL per second?

More ratios. Through the use of nuclear magnetic resonance spectroscopy, it is possible to determine the ratio between the protonated and deprotonated forms of buffers. (a) Suppose the ratio of [ A- ]A I to [HA] is determined to be 0.1 for a buffer with pKar6.0.pKa = 6.0. What is the pH? (b) For a different buffer, 91974 suppose the ratio of [ A- ]lA J to [HA] is determined to be 0.1 and the pHpH is 7.0. In this case, what is the pKapKa of the buffer? (c) For another buffer with pKa=7.5PKa = 7.5 at pH 8.0pH 8.0, what is the expected ratio of [ A- ][A ] to [HA]? do
