Chapter 25, Problem 49P
Interpretation Introduction

(a)

Interpretation:

Results in the graph are to be determined.

Concept introduction:

Lesch-Nyhan syndrome is an X-linked inherited disorder. It affects about 380,000 people across the globe. Due to this uric acid gets accumulated in the blood and urine, leading to hyperuricemia and hyperuricosuria. This also leads to gout and kidney problems.

Interpretation Introduction

(b)

Interpretation:

The reason for the Lasch-Nyhan syndrome in LND1 person is to be determined.

Catalase is the enzyme. experiment with h2O2 3. What is the marker for the enzyme’s optimum condition?
Need help   Sesamin is one of the major lignans found in sesame oil. Sesamin-metabolic pathways remain uncharacterized at both the enzyme and gene levels. However, one microorganism showed significant sesamin-degrading activity and was identified as Sinomonas sp. no. 22. A sesamin- metabolizing enzyme named SesA was purified from this strain and characterized. Explain how sesamin methylene transferase was identified and characterized and explain the mechanism. Based on your knowledge, what other assays could be run to characterize this enzyme?
The enzymatic activity of PFK1 is generally measured by set- ting up a coupled enzyme assay system whereby aldolase, triose phos- phate isomerase, and glycerol-3-phosphate dehydrogenase are added to the assay mixture. For the latter enzyme, NADH is added and its change in concentration is readily monitored at 340 nm. Write the chain of reactions catalyzed by these enzymes using structural formulas, label substrates and products, and explain why the coupled en- zyme assay system leads to oxidation of NADH. While the chain of reac- tions is similar to those in glycolysis, there is a critical difference because of the dehydrogenase enzyme. Describe how this enzyme causes the chain of reactions to differ from those in glycolysis.
