Chapter 7, Problem 46P

Antibodies were made that recognize six proteins that are part of a complex inside the Caenorhabditis elegans one-cell embryo. The mother produces proteins that are believed to assemble stepwise into a structure in the egg, beginning at the embryo’s inner surface. The antibodies were used to detect the protein location in embryos produced by mutant mothers [who are homozygous recessive for the gene(s) encoding each protein]. The C.elegans mothers are self-fertilizing hermaphrodites, so no wild-type copy of a gene will be introduced during fertilization.

In the following table, * means the protein was present and at the embryo surface, − means that the protein was not present, and + means that the protein was present but not at the embryo surface. Assume all mutations prevent production of the corresponding protein.

Chapter 7, Problem 46P, Antibodies were made that recognize six proteins that are part of a complex inside the , example 1

Complete the following figure, which shows the construction of the hypothetical protein complex, by writing the letter of the proper protein in each circle. The two proteins marked with arrowheads can assemble into the complex independently of each other, but both are needed for the addition of subsequent proteins to the complex.

The easily observed external structures in insects, such as different pairs of legs, antennae, and wings form from imaginal discs. a.) How is the identity of the different Drosophila imaginal discs established in the embryo—for example, how is it determined whether a disc will form a wing, a leg, or some other structure? b.) Dominant mutants in the gene Antennapedia result in fore legs forming in the location where antennae are expected. How does this mutant phenotype illustrate your response to Part a?
) Explain how and why dorsal/ventral polarity will be affected in fly Question 3 (1. embryos carrying the following mutations; also in each case darken in the area of the cells in the cross-sectional view of the fly embryo which are expected to express the paulie gene. D = dorsal; V = ventral. a) a mutation which results in the deletion of the cytoplasmic domain of the Cookie protein. b) A mutation which results in a constitutively active Bombe protein, i.e. the Bombe protein is always in an activated state. c) A mutation which causes the Pickle protein to be retained in the cytoplasm of the embryo.
The oncogenic protein BETA promotes entry into the S phase of the cell cycle. Phosphorylation of BETA at the amino acid Tyr98 causes BETA to be degraded by the proteasome, thus limiting its abundance. A mutation in the codon encoding Tyr98 changes this residue to Cys, which cannot be phosphorylated. What is the best description of this mutant allele?a) antimorphb) hypermorphc) hypomorphd) amorphe) neomorph
