Chapter 7, Problem 3Q
Summary Introduction

To review:

Evidence of signal amplification in the signal transduction pathway.

Introduction:

Different genes of a cell get activated in different circumstances. Bacterial cells like Escherichia coli express a high amount of OmpC in hypertonic solutions as compared to isotonic or hypotonic solutions. Other proteins, such as membrane protein (EnvZ), act as a protein kinase and get phosphorylated. They also phosphorylate proteins like OmpR (cytoplasm protein).

The amount of EnvZ and OmpR after incubation with 32P-radiolabeled ATP is recorded in the following table:

Table A

An isotonic solution consisting of nutrient broth only A hypertonic solution consisting of Nutrient broth and 20% sucrose solution
EnvZ 63 113
OmpR 2043 3525

Patch clamping can be used to measure the conductance properties of individual ion channels. Describe how patch clamping can be used to determine whether or not the gene coding for a putative K+ channel actually codes for a K+ or a Na+ channel.
What is signal transduction? Illustrate and describe the molecular events in signal transduction pathways involving 1) G-protein-coupled receptors and 2) enzyme linked receptors.
Explain, in detail, how tyrosine kinase proteins are involved in one signal transduction pathway of your choice. Make sure you describe the complete pathway in which it is involved. Then, describe how you would experimentally demonstrate the requirement of a tyrosine kinase protein in your chosen pathway.
