Chapter 4, Problem 4Q
Summary Introduction

To analyze:

The total GC (guanine and cytosine) and AT (adenine and thymine) content of deoxyribose nucleic acid (DNA) from the given data set and comparison of different organisms, on the basis of the combined AT and GC contents.

Given:

The percent (%) of DNA bases from some organisms have been compiled in the form of Table 1 below:

Table 1: Percent of DNA bases in some organisms.

Organism and the tissue from which DNA is extracted
Composition of DNA base (%)
Adenine Guanine Cytosine Thymine
Herring sperm 27.8 22.2 22.6 27.5
Rat bone marrow 28.6 21.4 21.5 28.4
Human sperm 30.7 19.3 18.8 31.2
Escherichia coli 26.0 24.9 25.2 23.9
Yeast 31.3 18.7 17.1 32.9

Introduction:

In every organism, the genetic material is passed from one generation to another generation in the form of a code, which is present in DNA. The DNA is a double-helix structure, which contains purines and pyrimidines. The purines present in DNA are adenine and guanine and the pyrimidines are cytosine and 5-methyl uracil (thymine).

Chargaff’s results yielded a molar ratio of 1.29 for A to G in ox DNA, 1.43 for T to C, 1.04 for A to T and 1.00 for G to C. Given these values, what are the approximate mole fractions of A, C, G, and T in ox DNA?
Assuming the average weight of a deoxynucleotide monophosphate (dNMP) is 327.0 g/mol, how many picomoles of DNA are present in 500ng of a 1000bp DNA fragment?
With regard to Chargaff’s experiment described in Figure shown, answer the following:A. What is the purpose of paper chromatography?B. Explain why it is necessary to remove the bases in order todetermine the base composition of DNA.C. Would Chargaff’s experiments have been convincing if theyhad been done on DNA from only one species? Discuss.
