Chapter 32, Problem 13P
Interpretation Introduction

Interpretation:

The configuration of reaction catalyzed by beta-galactosidase should be determined.

If each step proceeds with inversion of configuration, the overall stereochemistry of the mechanism should be determined.

Concept introduction:

Galactose and glucose are monosaccharide units, that undergo condensation reaction to produce lactose, a disaccharide. Further, hydrolysis of galactose by the enzyme lactase and beta-galactosidase yields back glucose and galactose. Beta-galactosidase is an enzyme which is produced by the lac operon in E. Coli.

Make an electron-flow-mechanism for this synthetic scheme. This involves predicting major and by-products using electronic and structural effects. The arrow push mechanism must be shown.(from the reaction of α-ketoacids and oxaprolines to proteins that contain native serine residues ) with label
The substitution of His 64 of carbonic anhydrase II with Ala results in a sharp decrease in the activity of the enzyme in HEPES buffer (molecular weight of HEPES = 238.3 g/mol). However, increasing concentrations of imidazole (molecular weight = 68.1 g/mol) restores the reaction rate close to that of the wild-type enzyme. Propose an explanation for these results.
Consider the role of Histidine in the Serine protease mechanism and sketch a plot showing the predicted pH profile of chymotrypsin which has a pH optimum of approximately ~8. The pk, for the His in the catalytic triad is 7.3 in free chymotrypsin which increases to greater than 8 with a bound peptide. Be sure to label the plot axes and indicate the pka of His on the plot,
