Chapter 22, Problem 4PDQ

One of the major causes of sickness, death, and economic loss in the cattle industry is Mannheimia haemolytiea, which causes bovine pasteurellosis, or shipping fever. Non invasive delivery of a vaccine using transgenic plants expressing immunogens would reduce labor costs and trauma to livestock. An early step toward developing an edible vaccine is to determine whether an injected version of an antigen (usually a derivative of the pathogen) is capable of stimulating the development of antibodies in a test organism. The following table assesses the ability of a transgenic portion of a toxin (Lkt) of M. haemolytiea to stimulate development of specific antibodies in rabbits.

* Lkt50 is a smaller derivative of Lkt that lacks all hydrophobic regions. + indicates at least 50 percent neutralization of toxicity of Lkt; − indicates no neutralization activity.

One strategy for vaccine development currently under investigation is the use of pathogen-derived T cell epitopes as a component of the vaccine. For viral pathogens, implementing this strategy involves scanning the predicted amino acid sequences of the viral proteins for likely peptide epitopes that would bind to MHC class I and MHC class II molecules. In addition to the complication of MHC sequence polymorphism in the human population, another complication of this strategy for peptide epitopes that would bind to MHC class II proteins is: The importance of viral proteins containing peptides that are cleaved into 8–10 amino acid long fragments. The ability of viruses to mutate their proteins to avoid MHC anchor residue sequences. The fact that long peptides (>13 amino acids) are rapidly degraded in cells. The fact that MHC class II proteins are intrinsically stable, even in the absence of binding to a peptide. The absence of defined sequence motifs that predict peptide binding to…
Two newly developed vaccine candidates (A and B) are tested in mice for their ability to elicit high concentrations of anti-meningococcal antibodies that would provide mucosal as well as bloodstream protection. Also, the ideal candidate vaccine should also provide long-lasting immunity to the infection. Below is a diagram of the results from the primary immunization with both candidate vaccines.  What is the predominant antibody isotype elicited by the primary immunization with these candidate vaccines? In which part of the body is that antibody primarily found?
There are currently 3 vaccines for COVID approved for use in the United States: Pfizer, Moderna, and Johnson & Johnson. Compare and contrast each in regards to the type of vaccine (mRNA or viral vector and what that means), how many doses they require (and the timing for those that require more than 1), and their efficacy.
