Concept explainers
Interpretation: The physiological effect predicted from a mutation that replaced with serine the cysteine in the constant part of immunoglobulin light chain and involved in disulfide-bond formation with the heavy chain should be explained.
Concept Introduction: There is a disulfide linkage between the cysteine amino acids that holds light chain in association with the heavy chain of the immunoglobulin.
Answer to Problem 1P
Solution: Because of the cysteine to serine mutation there will be no disulfide linkage between the light and heavy chain of the immunoglobulin.
Explanation of Solution
The cysteine helps in disulfide linkage formation between the light and heavy chain in the Fab fragment of the immunoglobulin. In the absence of cysteine, serine will not be able to form disulfide linkage between the light and heavy chain of the immunoglobulin. As a result of this mutation the light and heavy chain of the immunoglobulin will not stay physically connected to each other. This will cause the immunoglobulin to denature.
Thus, because of the cysteine to serine mutation there will be no disulfide linkage between the light and heavy chain of the immunoglobulin.
