Introduction:
Sanger’s method or dideoxy sequencing is a method of gene sequencing. It is also known by the name chain termination method. This method requires a single-stranded DNA template, DNA primer, DNA polymerase, dNTPs (deoxynucleotide triphosphates) and ddNTPs (di-deoxynucleotide triphosphates). In this method, 4 different ddNTPs (A, T, G, and C) are used and four separate reactions are carried out.
Gel electrophoresis is used for the separation and analysis of macromolecules and their fragments. Separation is done on the basis of size and charge. Gel electrophoresis is a laboratory technique mostly used to separate charged molecules like DNA and RNA. Ethidium bromide, fluorophores, and SYBR green are the staining dyes which are used to stain gels.
