Concept explainers
A process by which a polypeptide chain folds to form an active protein is known as protein folding. The structure of a protein is very critical for its function. A process by which a protein molecules move between cellular compartments is known as protein translocation. The translocation process advances the mRNA-tRNA moiety on the ribosome, in order to permit the next codon to enter into the decoding center. There are two translocation systems such as Sec system and Tat system and two secretion systems, such as Type I and Type IV are present in both Gram-positive bacteria and Gram-negative bacteria.
The translocation or secretion system that is most widespread of all the translocation system is the Sec system. The Sec system is occasionally termed as the general secretion pathway. This is because, it is highly conserved and have been identified in all three domains of life, such as eukaryotes, bacteria, and archaea. The Sec system translocates unfolded proteins across the plasma membrane or incorporates the unfolded proteins in the plasma membrane itself.

- Throughout downstream processing, various analytical methods must be used to evaluate the effectiveness of unit operations. Provide a method or methods (if more than one is required) for each scenario. Assess total protein in rice flour (solid material). Visualize antibody fragments (heavy chain and light chain) and compare molecular weights. Quick determination of the concentration of a purified sample of monoclonal antibody. Determine the concentration of monoclonal antibody in clarified extract. Study aggregation of a protein product without disruption of tertiary protein structure. Characterize phenolics (secondary metabolites and impurities produced by green plant tissue) by identifying phenolic species and relative concentrations. Detect protein variants that differ by a substitution of two amino acids. Quantify the level of endotoxin in the final purified product. Quantify bacterial contamination in your final formulated product.arrow_forwardWhat makes platinum replica EM more desirable than conventional SEM and TEM (address both, specifically) for imaging cytoskeleton?arrow_forwardDescribe the bioreactor design consideration to be taken in account during the bioprocessing of animal cell culture.
