In the synthesis of fatty acids, firstly acetyl thioester and malonyl thioester combines to form a four carbon thioester.  This thioester reacts with malonyl thioester.  Each malonyl thioester adds two carbon.  Each time malonyl thioester is added, two carbon atoms are increased in the fatty acid chain.
