Introduction:

Theta replication occurs in E. coli and other organisms that have circular DNA. E. coli contains circular DNA that contains “a single origin of replication” producing a “replication fork” is found. This replication can be both unidirectional and bidirectional. During theta replication, two strands of DNA strands get separated and replication of both the DNA strands occurs in a single or opposite direction. The circular replicon looks like “$\text{θ}$” (a Greek letter), thus this replication is termed as theta replication.

Introduction:

In a rolling circle method of DNA replication, a “unidirectional replication” of nucleic acid that synthesizes multiple copies of DNA or RNA rapidly. In this method, “the outer strand work as a positive (+) strand and the inner strand work as negative (-) strand”. DNA replication begins to start by creating a “nick (cut)” at the positive strand. The (+) strand has served as a template for the (-) strand. The (-) strands served as a template for making the (+) strand.
