Chapter 22.5, Problem 1COMQ
Summary Introduction

Introduction:

Shotgun sequencing is a method that is used for deoxyribonucleic acid (DNA) sequencing. Fred Sanger and his colleagues used shotgun sequencing for the first time to sequence the genome of microscopic organisms like, viruses and bacteria with small genetic material. The first organism that was sequenced using shotgun was Hemophilus influenzae. Craig Venter also used this method to sequence the human genome.

Shotgun sequencing is a method of DNA sequencing in whicha. the DNA fragments to be sequenced are randomly generatedfrom larger DNA fragments.b. the sequencing reactions are carried out in rapidsuccession.c. the samples to be sequenced are rapidly generatedby PCR.d. all of the above occur.
When we discuss PCR and other similar techniques, the term Tm is often used. This refers to a.The temperature where the DNA molecules denature.  b.The temperature of the first step in a PCR cycle. c.The temperature where half of the DNA molecules are denatured. d.Tm is the same as the annealing temperature.  e.The Temperature at which all of the DNA molecules are dentatured
Which of the following is true?   a. In one technique, the DNA sequence can be determined using one strand of DNA as it passes through a nanopore.   b. Next generation sequencing is slow but accurate.   c. Both of the above are true.
