Chapter 10.4, Problem 5CC
Summary Introduction

To find: The role of glyceraldehyde 3-phosphate (G3P) in glycolysis and Calvin cycle.

Introduction:

Glycolysis is a stepwise process by which one molecule of glucose breaks into two molecules of pyruvic acid by partially oxidizing glucose. This process occurs in the cytoplasm of the cell.

Calvin cycle is a biochemical pathway for the reduction of CO2 or photosynthetic carbon. The acceptor molecule is a 5-carbon ketoses sugar i.e. Ribulose 5-bisphosphate (5 RuBP).

. Consider the enzymatic reaction scheme: Asparagine + H20 Aspartate + NH3: a) calculate the specific activity of the enzyme, if in 30 seconds as a result of a reaction involving 3 mg of the enzyme under optimal conditions (pH 8.0, 37 °C) 75 umol aspartate is obtained; b) describe the reasons for the decrease in enzyme activity after incubation for 6. 10 minutes at 70 °C (provide an appropriate graph).
Suppose that your Ph.D. mentor asks you to compare the kinetic behavior of the two recently discovered peptidase isozymes PepA and PepB. Your mentor's initial studies identified a pentapeptide substrate called Tide5 that appeared to be able to distinguish the kinetic behaviors of PepA and PepB. All they had time to tell you was this: At low Tide5 concentration, the ?o (initial velocity) of PepA was greater than PepB, but at high Tide5 concentration, the reverse was true. Before you start your kinetic assays, you decide it would be wise to ponder this small bit of information. You have no numbers to work with, but does it tell you something about the relative values of ?m and ?max for PepA and PepB? Select the statements that are consistent with the information that you were given.
