Chapter 53.2, Problem 3C
Summary Introduction

To sketch: The graphs representing the long-term growth of two populations of bacteria cultured in test tubes, one in which the nutrient medium is replenished and the other in which it is not replenished.

Introduction: Bacterial growth very much depends on the availability of nutrients in the growth media. The concentrations that are higher or lower than the optimum levels can affect the growth of the bacteria in the media. It is very important to replenish the nutrient media timely to ensure continuous and healthy bacterial growth in the media.

There are two cultures of yeast cells in the pictures, one has been incubated for 6 hours and one has been incubated for 24 hours. After a 10x dilution by taking 100µl of each culture and adding it to 900 µl water in a microcentrifuge tube and 100µl sample from the tube was taken to view in the counting chamber. a) Count the total number of yeast cells for each culture respectively b) Calculate the concentration and density of yeast cells for each culture respectively
xplain the four stages of the growth curve for a microorganism.  What is the generation time? Which aspect of the growth curve is used to calculate the generation time and why?  How is the generation time calculated and why does it make sense that E.coli and M.tuberculosis have such drastically different generation times?
Use the following passage to answer 2 consecutive questions. A urine specimen was collected in a container and placed on the central desk for delivery at 10:00 am. The specimen is not sent to the lab until 5 hours later (3:00 pm). The sample time was marked by the central clerk as collected at 4:00 pm. The bacterial species has a generation time of 30 minutes at room temperature. There are 100 bacterial cells/ml at the time of collection (10:00 am). The laboratory will cite a bacterial infection at more than 1000 bacterial cells/ml.
