Concept introduction: The chain termination method or Sanger sequencing method was devised by Frederick Sanger in 1977. In this technique, the template DNA to be sequenced is incubated in a tube with DNA polymerase, a suitable primer, and the four DNA nucleotides (dATP, dTTP, dGTP, and dCTP) for the polymerization reaction. The dye labeled, chain-terminating dideoxynucleoside triphosphates (ddNTP) that lack the 3ʹ-OH group of deoxynucleotides are added in small amount to the reaction sample. The addition of next nucleotide for chain growth requires a free 3ʹ-OH group; hence, addition of ddNTP terminates the chain elongation. This process is repeated in a number of cycles to obtain different fragments of DNA.
