Chapter 24.2, Problem 1TQ
Summary Introduction

To review:

The ability of antibody fragment F(ab’)2 to prevent the binding of rhinovirus with ICAM-1 receptors on the surface of the host cell and the ability of antibody fragment F(ab’)2 to facilitate microbial phagocytosis.

Introduction:

The antibody is comprised of two arms that are heavy and light chains. The heavy chains are attached by disulfide bonds. The antibodies have two regions, constant and variable regions. ICAM-1 or intercellular adhesion molecule 1 is one of the proteins encoded by gene ICAM-1. This gene encodes the glycoprotein present on the cell surface that expresses itself on the cell of the endothelium and immune system. It is one of the transmembrane protein and a member of a superfamily of immunoglobulins.

Which of the following is false when considering the CCR5Δ32 mutation? a) The mutation prevents the entry of HIV-1 into a T cell. b) The deletion that produced CCR5Δ32 caused a frameshift which extends the length of the protein product. c) CCR5Δ32 prevents infection by HIV-1 in homozygotes but only delays the onset of HIV symptoms in heterozygotes. d) The protein product produced by CCR5Δ32 is altered in such a manner that it cannot embed in the cell membrane.

Questions related to SARS-CoV-2 Inhibitor Science Article (Zhang et al.) 1) What is the function of the viral main protease? 2) What is the amino acid recognition sequence that is cleave by this main protease? Indicate where this protease cuts? 3) Would interfering with this protease effect other proteases in human cells? 4) How closely related are the amino acid sequences between SARS-CoV-2 and SARS-CoV? What does this suggest about focusing research on inhibiting this enzyme?
What is the purpose of blocking the Western Blot membrane? A) To cover the membrane with a solution of proteins that serve as a substrate for secondary antibody binding B) To maintain target proteins in their native, non-denatured state C) To cover the membrane with a solution of proteins in order to reduce non-specific binding of the antibody to the membrane D) To enhance non-specific background staining through interactions with the proteins present in the blocking solution
