The predominant form of proline at physiological pH 7.4 has to be identified.

• Identify ${\text{pK}}_{\text{a}}$ of the carboxylic acid group and determine which form predominates.
• Identify of $\text{α-amino}\text{group}$ and determine which form predominates.
• Identify ${\text{pK}}_{\text{a}}$ of the side chain, if necessary, and determine which form predominates.
• ${\text{pK}}_{\text{a1}}$ refers the acidity of the carboxylic acid group, and then ${\text{pK}}_{\text{a2}}$ refers to the acidity of ammonium group. ${\text{pK}}_{\text{a3}}$ Refers to the acidity of side chain containing either acidic or basic groups present in the amino acid.
