Interpretation:

Of the restriction enzymes listed in the table, the enzymes that generate flush, or blunt-ended, fragments should be identified. The cuts that cannot be converted to flush ends by the action of DNA polymerase should be identified and explained.

Concept introduction:

Many restriction enzymes create a double-stranded cut between the recognition site which results in a flush end or blunt end. Examples of blunt end cutters are AluI and PvuII. Some of the enzymes cleave the two DNA strands in such a way that they are not cleaved at the exact same position. Rather cleavage is of staggered type by 2 or 4 nucleotides which results in DNA fragments having short and single-stranded protrudes at both the ends of DNA and are known as sticky ends or cohesive ends as base pairing in them has the capability to stick the DNA molecules again back together.
