An antibiotic is a type of antimicrobial substance active against bacteria. They are the most important commercially used secondary metabolites, produced by many soil microorganisms like bacteria and fungi, and employed in a wide range. The most important antibiotics used today are of microbial origin.
Screening of antibiotic-producing organisms are isolated from soil by various steps like the collection of the soil sample, preparation of soil sample, isolation of microorganisms, inoculating the sample, subculturing the particular organism, stain characterization, biochemical characterization.
Screening of antimicrobial activity for antibiotic production is done by adding Mueller Hinton agar media was prepared by adding 17 g of MHA media in 500 ml of distilled water and autoclaved at 121°C for 20 minutes. After sterilization, the media was cooled and poured into Petri dishes and kept in an incubator at 37 °C for 24 h to check its sterility. Two test tubes were taken containing 2 ml of sterilized nutrient broth.
