The most significant weakness of our experiment was that the repeats were conducted on different days. Because the repeats were done on different days and at different times, the environment in which they were conducted wasn’t the same. This meant that environmental factors such as temperature could have affected the experiment and its outcome. If it had been warmer, then there would have been less CO2 dissolved in the water, which would have resulted in less H2CO3. If the repeats were conducted in a cooler temperature than the previous repeats, then it could have meant that there was more CO2 dissolved in the water, so more H2CO3 would have been produced. In Lab 3.2, we burned for different chemicals and each produced a different color. The colors were different because each element, when exited, gain more energy and when the electron release that energy and jump to a more stable level/orbit, it produces a specific color that corresponds with specific wavelength that matches with each different element. Since chemicals have certain colors and wavelengths, when different kinds of chemicals are burned, you can learn what is in that chemical depending on what is produced. If a certain chemical has several blue wavelengths and only a few red wavelength, the chemical will burn blue because the blue wavelength are stronger than the red. The red wavelength will still be there but can not be seen.

Data were collected in September and November 2017 at the Elk Valley Preserve and Field Station in Banner Elk, NC. The preserve is located at 36°10'17.2"N 81°54'45.9"W, in Avery County in western North Carolina. The area is a mixed deciduous forest at an altitude of 1,127 m above sea level. Data were collected in early fall, during which temperatures average around 10-18 degrees Celsius. Summers in Banner Elk are typically mild, with temperatures averaging 21 degrees Celsius, and winters are cold and snowy, averaging near zero degrees Celsius. During the months of September and November there is an overall average precipitation of 12-25cm.

Water was of Milli-Q quality (Millipore, Bedford, NY). Hypo-cotyledons of Sesuvium portulacastrum were used as ex-plant for initiation of callus cultures.
Silibinin from Sigma-Aldrich (St Louis, MO, USA), terbium (III) chloride hexahydrate (TbCl3.6H2O) from Acros Organics (New Jersey, USA), 2-amino-2hydroxymethyl-propane-1,3-diol-hydrochloric acid (Tris-HCl) and hydrochloric acid and methanol from Scharlau (Barcelona, Spain) was purchased. Deionized distilled water was provided from Shahid Ghazi Pharmaceutical Company (Tabriz, Iran). 1000 mg/L of silibinin stock standard solution was prepared in methanol after sonication for 10 min. The working solutions were prepared daily with appropriate dilution with water. Terbium (III) chloride hexahydrate stock solution (5× 10-2 M) was prepared by dissolving 0.465 g of TbCl3.6H2O in 25 ml water and diluted by distilled water to prepare working solutions.

Liquid medium was exploited to scrutinize the color strength measurement of the Remazol Black B decolorization activity by dexterous bacterial isolates. Based on the comparative ability of the decolorization of different isolates, three best performing isolates ( 1, 2 & 3) with more than 90% of decolorizing activity were chosen for further experiments (Data not shown). Biodecolorization of Remazol Black-B by selected bacterial isolates Biodecolorization of Remazol Black-B by the selective bacterial isolates (ETL-A, ETL-B & ETL-C) was confirmed by conducting another experiment in liquid medium at different time periods (Figure 1). It was found that different bacterial isolates had variable potential to remove Remazol Black-B in the growing cultures. The most efficient bacterial isolate to decolorize the Remazol black-B was ETL-A with 98% color removal efficiency in 18 h incubation period while remaining isolates displayed maximum decolorization in 24 h. The bacteria was isolated from six different types of mango fruit (Mangifera sp.) that were Gedong Gincu mango (A), Budi Raja mango (B), Gedong mango (C), Dermayu mango (J), Harum Manis mango (K), and Simanalagi mango (L), two mangos for each type. We took two samples, 10 grams each, from the upper part and the below part of each mango, and that made a total of 24 samples. The flesh was smashed and mixed with 90 mL of MRSB (de Mann Rogosa Sharpe Broth), then incubated for 24 hours on 37oC. 1 ml of the sample then diluted with physiological solution; for mango A, B, and C diluted in 10-1 and 10-2, for mango J, K, and L diluted in 10-2, 10-3, 10-4.
Studies show that unwashed or lightly rinsed vegetables can harbor pathogenic bacteria and have been implicated in numerous foodborne infections. The objective of this experiment was to obtain a quantitative viable plate count of enteric bacteria present in a sample of broccoli sprout water. To enumerate the number of bacterial colonies present in the sample, we performed serial dilutions of broccoli water spread onto MacConkey Agar plates. MacConkey Agar is a selective and differential media. It is used to select enteric bacteria and differentiate lactose-fermenters from non-lactose fermenters.
