I received 2.5 out of 3 possible points for this question. Based on the TA’s notes, it seems as though I lost ½ of a point for not saying that “more ddNTPs will result in shorter sequences”. However, in my answer, I did in fact say that increasing the ratio of ddNTPs to dNTPS will cause DNA sequencing to stop quickly, namely at the first few nucleotides if many ddNTPs are added. Because I included such information in my answer, and given the accuracy of the rest of my answer, I’m not sure why I lost ½ of a point and instead believe that I did not deserve to lose any points, or at least did not deserve to lose ½ of a point. I received 2.5 out of 3 possible points on this question. First, as I approached this question, I remembered our discussion in class of methods for determining which stage of the cell cycle cells are in, and how doing so indicates growth. In class, you mentioned that cell size can be used to determine which stage of the cell cycle certain cells are in. Although you said that measuring the amount of DNA is a more reliable method, you nonetheless said that cell size can be used for this. Some of the answers on the answer key are methods used to determine which stage cells are in, as doing so can indicate whether or not cells are growing. This was the case for 2 of my correct answers (for example, I mentioned using formation of the mitotic spindle as a way to measure cell growth, as doing so measures a cell's’ entrance into M-phase, which inherently indicates cell growth). So, I am not sure why measuring cell size is not considered as a sufficient answer for part of this question. Q8. I received 2.5 out of 3 possible points for this question. For the first part of the lab, our goal was to calculate the time constant, , of an RC circuit. We made an RC series circuit and connected it to the Rigol wave generator to produce a square waveform for current. Then, we collected data of the voltage across the capacitor at different points in time using a myDAQ and the 4BL application. In order to find the time constant, we linearized the voltage we measured across the capacitor and then performed a linear regression on the data. The goal of this lab was to expand and centralize our infrastructure by adding a Windows Server 2012 VM and configuring it with a number of key services. Some of the services that we deployed on the Windows 2012 VM had previously been configured on other machines, namely the RHEL VM from Lab 2. We began by installing and configuring the server itself, ensuring that we provided it with ample memory and processing power. Next, we configured DHCP, disabling the DHCP service running on our RHEL VM. We then continued by installing Active Directory and DNS on the Windows 2012 VM, disabling the DNS server running on the RHEL VM in the process. The DNS server on RHEL would later be reactivated as a secondary (slave) server. We then proceeded to set up monitoring for both DHCP and DNS.
