Replication occurs but as soon as the dideoxynucleotide analog of the nucleotides is confronted the replication stops. But since the concentration of dideoxynucleotide is low replication halts at specific locations and thus yields fragments of different sizes. The fragments are then analyzed by gel electrophoresis.
