To study bacteria and other microorganisms, it is necessary to grow them in controlled conditions in the laboratory. Growth media contain a variety of nutrients necessary to sustain the growth of microorganisms. There are two commonly used physical forms of growth media: liquid media and solid growth media. A liquid medium is called a broth.
A growth medium can be both selective and differential. For example, EMB (eosin methylene blue agar) inhibits the growth of Gram positive bacteria. Gram negative bacteria that grow on this medium are differentiated based on their ability to ferment the sugars lactose and sucrose.
- Media that inhibit the growth of unwanted microorganisms and support the growth of the organism of interest are called selective media. Selective mediums contain particular ingredients that inhibit the growth of certain microbes. An example of a selective medium is MacConkey agar.
- Differential media are bacteriological growth media that contain specific ingredients to allow one to distinguish selected species or categories of bacteria by visual observation. Differential Media are used to distinguish between closely related organisms or groups of organisms. One example of a differential medium is blood agar. Blood agar distinguishes microbes based on their ability to lyse red blood cells (RBCs), a property known as hemolysis.
