The three main characters are Daniel Burnham, H.H. Holmes (Herman Webster Mudgett), and Frank Geyer. The first protagonist is Daniel Burnham, an architect who, along with his partner John Root, is tasked to construct the 1893 World's Fair. Throughout the construction of the World's Fair, Burnham has to overcome many incidents that prevent him from completing the fair. The second protagonist is Detective Frank Geyer, a "big man with a pleasant, earnest face" (Larson, 339). Geyer is brought onto an insurance fraud case, which shortly escalates to a murder case against the antagonist, H.H. Holmes. He a psychoctic doctor from New Hampshire. Holmes is a merciless killer, with persuasion skills capable of even subverting the police.

The Devil in the White City takes place in Chicago, the "White City", during the early 1890s. With Chicago winning the location for the 1893 World's Fair, people from all over the United States and abroad travel to the White City.
