Step 1: Chain termination method

This method is used for DNA sequencing and it relies on the use of Deoxyribonucleoside triphosphates, derivative of normal triphosphates that lack the 3' hydroxyl group. 

To determine the complete sequence of a DNA fragment, the double-stranded DNA is first separated into single strand and the strand with 3'-5' direction acts as a template strand. The bands on the gel are read in the order starting at the bottom of the gel and working across all lanes, the DNA sequence of the newly synhesized strand can be determined. 

The band on the gel appear with respect to their size as we move down to the lane the band size decreases.
